Setting pre-build markup

With pre-builds, is it possible to set the sell price or sell price % markup?
I have tried to update 'TotalEx' on a standard price pre-build, the call succeeds and yet the price doesn't change. I have also tried this after un-checking 'Based on Defaults' in the UI and saving.

Unfortunately, for now only Total amount can be updated for a pre-build item.

I am trying to update TotalEx for a prebuild. Is this what you meant?
My request is

PATCH {{SimPRO:Api:Uri}}/api/v1.0/companies/2/prebuilds/setPrice/{{preBuildId}}
Authorization: Bearer {{bearerToken}}
Content-Type: application/json
Accept: application/json
{

}
and I am getting a response with a status of 204(Not content).
Any help would be appreciated.

204 means your request was successfully fulfilled. You could send a GET request to confirm the new TotalEx price after that.
